Cytogenetic Analysis: C-33 A細胞This a pseudodiploid human cell line with the modal chromosome number of 46, occurring in 70% of cells examined. Polyploid cells occurred at 8.6%. Seven marker chromosomes were consistently detected per pseudodiploid cell. They are: t(1q17q), t(1p21q), del(18) (q21.3), der(1)t(1;17) (p16;q21.3) and three others. Several other markers were also found but they occurred only once in 15 metaphases analyzed. Neither DMs nor HSRs were detected. Structurally normal N1 was absent. Generally there are two X chromosomes in each cell.

Comments: The C-33 A cell line is one of a series of lines (see also ATCC CRL-1594 and ATCC CRL-1595) derived by N. Auersperg from cervical cancer biopsies. The line exhibited a hypodiploid karyotype initially, and an epithelial morphology. Karyological instability was observed with continued passage. The retinoblastoma protein (pRB) is present but abnormal in size. Expression of p53 is elevated, and there is a point mutation at codon 273 resulting in a Arg -> Cys substitution. The cells are negative for human papillomavirus DNA and RNA.

Propagation: C-33 A細胞ATCC complete growth medium: The base medium for this cell line is ATCC-formulated Eagle's Minimum Essential Medium, Catalog No. 30-2003. To make the complete growth medium, add the following components to the base medium: fetal bovine serum to a final concentration of 10%.

Temperature: 37.0°C

Atmosphere: air, 95%; carbon dioxide (CO2), 5%

Subculturing: Protocol: Remove medium, and rinse with 0.25% trypsin, 0.03% EDTA solution. Remove the solution and add an additional 1 to 2 ml of trypsin-EDTA solution. Allow the flask to sit at room temperature (or at 37C) until the cells detach. Add fresh culture medium, aspirate and dispense into new culture flasks.

Subcultivation Ratio: A subcultivation ratio of 1:3 to 1:8 is recommended
