GAS-GFP報告基因是自主研發的用于檢測STAT1/STAT1轉錄活性水平為目的的報告基因。GAS(gamma-activated sequence, GAS) 又稱γ干擾素活化序列在皮膚發生遲發型接觸性過敏時,可與轉位至角朊細胞核內的STAT-91結合而激活K17的表達。
GAS-GFP報告基因主要用于檢測細胞中干擾素調控的信號通路的活性、**研究以及基因過表達和RNAi的表型分析等。
pGMGAS-GFP是改造后的哺乳動物真核表達載體,在其多克隆位點插入了多個STAT1/STAT1結合位點,可以高效地檢測STAT1/STAT1的激活水平。由于載體采用了GFP作為報告基因,更便于后續的檢測。同時,對載體中預測出的其它轉錄因子以外的結合位點進行了適當的突變,增加了質粒的轉錄因子結合特異性。另外,由于質粒體積減小,使得GAS-GFP報告基因更易于轉染。GAS GFP Reporter Plasmid(GAS-GFP報告基因質粒)
