HNF4熒光素酶報告基因（報告基因質(zhì)粒）（HNF4 luciferase reporter plasimd）是自主研發(fā)的用于檢測HNF4轉(zhuǎn)錄活性水平為目的的報告基因。HNF4(Hepatocyte Nuclear Factor 4)是轉(zhuǎn)錄因子核受體超家族中的一員，參與肝細胞分化調(diào)控和維持肝細胞的生物學功能。HNF4α主要在消化系統(tǒng)、肝臟和腎臟表達，以及在白色脂肪組織和胰腺中也有少量的表達。HNF4α具有阻斷肝纖維化、肝硬化、肝癌的**進程,改善肝臟功能的作用。另外,HNF4α在肝干細胞移植方面也發(fā)揮重要作用,提高肝細胞移植的成功率。

HNF4報告基因主要用于檢測細胞中HNF4的轉(zhuǎn)錄活性、**研究以及基因過表達和RNAi的表型分析等。

pGMHNF4-Lu是改造后的哺乳動物真核表達載體，在其多克隆位點插入了多個HNF4結合位點，可以高靈敏度地檢測HNF4的激活水平。同時，對載體中預測出的其它轉(zhuǎn)錄因子以外的結合位點進行了適當?shù)耐蛔儯黾恿速|(zhì)粒的轉(zhuǎn)錄因子結合特異性。由于質(zhì)粒體積減小，使得HNF4報告基因質(zhì)粒更易于轉(zhuǎn)染。HNF4 luciferase reporter plasmid （HNF4-Luc熒光素酶報告基因質(zhì)粒）

使用說明
